Sermorelin (GRF 1-29 NH2) is a synthetic analog of the first 29 amino acids of endogenous growth hormone-releasing hormone (GHRH). It stimulates the pituitary somatotroph cells to produce and release GH in a physiological pulsatile pattern, preserving the natural GH-IGF-1 axis feedback mechanism. It was FDA-approved for pediatric GH deficiency and is widely studied for adult hormone optimization.
Sermorelin binds GHRH receptors on anterior pituitary somatotrophs, activating adenylyl cyclase, increasing cAMP, and triggering GH synthesis and release. Unlike direct GH administration, sermorelin preserves the hypothalamic-pituitary feedback loop — GH and IGF-1 still suppress further release, preventing supraphysiological levels. This results in a more natural GH pulse pattern.
